Canine longevity research accelerates as Dog Aging Project studies 50,000 companion dogs

Researchers and companies are pushing a new phase of canine longevity work, building on the Dog Aging Project’s study of 50,000 companion dogs to test whether treatments can extend healthy life in pets and, potentially, people.

The Dog Aging Project began in 2014 and has grown into a large community-driven resource that researchers say can reveal which interventions might slow ageing. The effort has prompted clinical trials and commercial development aimed at extending healthspan rather than addressing single diseases. A growing trend among dog owners shows an increased commitment to their pets’ health, with lifetime care costs rising significantly, influenced by higher spending on health insurance and wellness plans.

Prof Matt Kaeberlein of the Dog Aging Project said the shift in ambition was obvious once researchers could combine biology and intervention testing. “I think what really got me excited was realising that we could not only understand the biology of ageing in dogs, but also apply interventions that have already been shown to slow ageing, extend lifespan and improve healthspan in mice,” he said. Studies suggest that canine aging research not only benefits dogs but also enhances our understanding of human aging, given the shared biological traits and environments. That work is moving into formal trials. A clinical trial of rapamycin is listed for 2026, and researchers have previously identified a group of interventions they judge safe enough to test in companion animals. “There are 8–10 interventions that are safe enough that they can and should be tested in dogs,” Prof Matt Kaeberlein said. Currently, the off-label use of rapamycin is gaining attention for its potential anti-aging benefits, focusing on improved heart health, mobility, and lifespan through enhanced cellular energy use.

Researchers are not the only actors. Loyal, a biotech company developing drugs for canine longevity, has advanced an experimental compound and received regulatory clearance for its programme: the US Food and Drug Administration approved Loyal’s experimental drug LOY-002, according to the organisations involved. “Our clinical trial is unlike any other in the sense that we’re not focused on treating a specific disease or ailment, but on extending healthy lifespan overall,” said Celine Halioua, Founder and CEO of Loyal. This aligns with a broader industry movement where pet healthcare spending is on the rise, reflecting owners’ commitment to ensuring their dogs enjoy longer, healthier lives.

Prof Daniel Promislow, also affiliated with the Dog Aging Project, and Dr Rachel Melvin are among the named researchers associated with the broader effort to map ageing in pet dogs.

Clinical work with rapamycin — an immunosuppressant repurposed at low doses for ageing research — has already shown benefits in controlled studies, and researchers are organising larger trials to measure effects on heart function, cognition and mobility as well as overall survival. Prof Matt Kaeberlein said he judged it better than even odds that rapamycin will increase lifespan and improve health in dogs: “I think it’s a better than 50/50 proposition that rapamycin increases lifespan and improves health in dogs.” The compressed lifespan of dogs, compared with humans, makes them a valuable model for such studies.

Funding and policy shifts have punctuated the project’s trajectory: a funding cuts announcement occurred in 2023, and teams have continued to expand enrolment and trial capacity. The scale of the Dog Aging Project — tens of thousands of enrolled animals — lets scientists test questions about how lifestyle, genetics and treatments interact across breeds and environments.

Individual animals have been used to illustrate differences in ageing across dog types. The research also highlights wide variation in breed lifespans: some breeds, such as the presa canario, average about 7.7 years, while others, like the miniature dachshund, can live nearly twice as long. As a reflection of these trends, dog owners are increasingly prioritizing veterinary care.

Scientists argue the compressed lifespan of dogs, compared with humans, and shared environments make them a useful translational model. That argument underpins both public-academic work and private-sector trials, including those run by Loyal and by university-linked teams.
